  • The correlation between the expression of plasma exosomes LncRNA MIR155HG and the severity of atopic dermatitis in children

    Objective To detect the expression of long chain non-coding RNA(LncRNA) MIR155HG in plasma exosomes in children with atopic dermatitis(AD) and analyze its relationship with the severity of the disease. Methods From December 2021 to January 2023, 118 children with AD in the Dermatology of Hebei Children's Hospital were selected as the AD group, and 120 healthy children in the same period were selected as the healthy control group. Isolate and identify plasma exosomes from the tested children, and detect the expression level of plasma exosomes LncRNA MIR155HG. Compare the expression levels of plasma extracellular vesicle LncRNA MIR155HG and the score of AD score(SCORAD) in children with different degrees of AD. Pearson method was used to analyze the correlation between plasma extracellular vesicle LncRNA MIR155HG and SCORAD score in children with AD. The diagnostic value of analyzing the expression level of LncRNA MIR155HG in plasma exosomes using the receiver operating characteristic curve(ROC) for moderate to severe AD. Results Compared with the healthy control group, the expression level of plasma extracellular vesicle LncRNA MIR155HG in the AD group was significantly increased(t/P=23.843/<0.001). The expression level of plasma extracellular vesicle LncRNA MIR155HG and SCORAD score in mild, moderate, and severe subgroups of AD patients increased sequentially(F/P=41.773/<0.001, 260.201/<0.001). The expression level of plasma extracellular vesicle LncRNA MIR155HG in children with AD was positively correlated with SCORAD score(r/P=0.673/<0.001). The ROC curve showed that the area under the curve(AUC) for diagnosing moderate to severe AD was 0.831 and 0.802, respectively, based on the expression level of LncRNA MIR155HG in plasma exosomes. There was no statistically significant difference in AUC diagnosis between moderate and severe AD(Z/P=1.008/0.313). Conclusions The high expression of plasma extracellular vesicle LncRNA MIR155HG in children with Alzheimer's disease may be related to the severity of the disease, and has high diagnostic value for the severity of AD. Clinical monitoring of changes in plasma extracellular vesicle LncRNA MIR155HG levels may be of great significance for targeted prevention and treatment of AD in children.
  • Cdc42 protein participates in neutrophil Elastase induced Mucoprotein hypersecretion in human airway epithelial cells

    Objective To investigate the relationship between cell division Cyclin 42(Cdc42) and neutrophil Elastase(NE) induced hypersecretion of Mucoprotein(MUC) 5AC in airway epithelial cells. Methods Human airway epithelial cells(16HBE) were cultured in vitro and intervened with Cdc42 siRNA and NE. The cells were divided into six groups: control group, NE group, NE+Cdc42 siRNA group, Cdc42 siRNA group, negative siRNA group, and NE+negative siRNA group. Western blot method was used to detect the relative content of Cdc42 protein, and ELISA and immunofluorescence were used to detect the expression level of MUC5AC protein in each group of cells. Results Compared with the control group, the content of Cdc42 protein in the NE group was significantly increased(P<0.05), and the expression level of MUC5AC protein in cells was also significantly increased(P<0.001); Compared with the NE group, the MUC5AC protein level in the NE+Cdc42 siRNA group was significantly reduced(P<0.05), and the Cdc42 protein content was also significantly reduced(P<0.05); The MUC5AC protein content in the Cdc42 siRNA group and the negative siRNA group showed little change compared to the control group(P>0.05). Conclusion Cdc42 may play an important mediating role in the formation of NE induced mucus hypersecretion in airway epithelial cells.

  • Progress in the Mechanism of Obesity Mediated Atrial Fibrillation

    Obesity has adverse effects on cardiovascular hemodynamics and cardiac structure, and increases the incidence of atrial fibrillation. Obesity leads to the accumulation of myocardial outer membrane fat and is associated with left atrial electrolytic remodeling. This article summarizes the mechanism of obesity mediated atrial fibrillation and the adverse prognosis of obesity in patients with atrial fibrillation through existing evidence.
